10X Chromium Single Cell sample submission and requirements: Due to the COVID-19, there are some changes regarding sample submission for 10x single cell. Please read below carefully and make sure you follow the Yale EHS guideline regarding the social distancing and wear a mask while submitting samples. Before you start your project: 1. Check 10X Genomic website, its support pages, especially the cell preparation pages for the buffers used for single cell suspension. 2. Contact us and arrange a meeting to discuss your project if possible 3. Familiar with the cell preparation protocol for your project; go through a mock run for cell preparation. Counting the cells and check the viability. Schedule for your project 1. Please arrange a time and date for your 10X experiments with Dr. Guilin Wang, at least a week of time if possible and send a calendar invite. Please be specific with the assay you would like to perform (5P, 3P, TCR/BCR, feature barcoding, etc.) so that we make sure we have the right reagent kits for you. 2. Due to the time sensitive nature of the 10x single cell sample prep, please make sure you can come at the arranged time and date to Room 2127, 300 George St. 3. You should have counted your cells and adjusted to about 1000 cells/ul (700- 1200) with the viability at least 70% (see below for more information). On the day of your scheduled time 1. Please remind us when you start your cell preparation and keep us posted. 2. Prepare your cells and adjust the concentration as following: Minimum concentration of 200 cells/ul (700 to 1200 cells/ul optimal range) in a volume of at least 30ul if possible. Please count your cells in your lab before come to us. We would not check your cells before loading on the chip. If possible, please provide 70ul of single cell suspension (two attempts at chip loading in case of clog). Single cell suspension should be at least 70% live (Trypan Blue stained) and free of visible debris and doublets. Recommend cell suspension buffer is PBS/0.04% BSA. Cell suspension buffers should be free of EDTA and Mg++ as well as free of DNase to be compatible with single-cell assay. Please see 10X cell preparation guide with more buffer choices. For fresh samples submitting 10-40K cells total is best. For frozen samples, 100,00 cells are the minimum, but 1 million are better. In case that you don’t have plenty of cells and want to use all you have without counting, you will be responsible for any outcome of the assay. Please let us know the number of cells targeted and the type of assay (single cell 3’ or 5’, TCR/BCR, feature barcoding, ATAC-seq, etc) Record the sample information in the sample submission form and bring the form with your samples. 3. Give us at least 45 min ahead of time when you are ready, so that we can start thawing the reagents and getting ready for the assay before you come. 4. Bring your cells on ice with your sample submission form and drop them off at the front door of room 2127 at 300 George St at the scheduled time. We would pick them up once you leave to avoid the in-person contact. We will wait for 30 minutes the most at the arranged time. Any delay longer than 30 minutes might void the arrangement. 5. We would load your cells according the information on the sample submission form. We would not check your cells before loading. Make sure the concentration and the viability are accurate so that you would capture the number of targeted cells. 6. Stay safe and be well
